The PhD student will be involved in the project "Hedgehog signaling in
the adult pituitary gland and in pituitary adenoma formation". The
project is funded by the Deutsche Forschungsgemeinschaft e. V. The
project can be started in February 2017.
The Hedgehog (HH) signaling pathway plays a crucial role during the
development of the pituitary gland which represents a gland with
superior function in the neuroendocrine system. We recently showed
that activation of HH signaling results in enhanced proliferation of adult
Sox2+ and Sox9+ pituitary stem cells and in elevated expression of
pituitary hormones (Acth (adrenocorticotropic hormon), Gh (growth
hormone), Prl (prolaktin). Together with our findings that human
pituitary adenoma show excess activation of the HH signaling cascade
these data might open new possibilities for novel targeted therapies
against hormone-producing adenoma and associated diseases (e.g.
Morbus Cushing, acromegaly and hyperprolactinemia). The current
project aims to validate the role of HH signalling in the homoeostasis of
the adult pituitary gland and in pituitary adenoma formation. Therefore
mouse models for lineage tracing of Gli1 expressing pituitary cells and
for the conditional overactivation of HH signaling in the adult pituitary
gland will be generated. Additionally human pituitary adenoma cultures
it will be used to analyze if the overactivation of the HH signaling
pathway is indeed involved in the formation and/or hormone production
of PA and thereby might be the target for targeted therapies.
Qualifications and experience:
you hold a master or diploma degree in natural science
you have experience with cell culture techniques and with
methods used in molecular biology
experience with murine (tumor) models and flow cytometry will
you are interested in aspects of signal transduction, tumor
genetics and the development of new tumor therapies
you have good IT and communication skills in English
you are team and goal-oriented
