Project description:
The group "Intestinal Barrier Function and Mucosal Healing" is interested in the pathophysiology of inflammatory bowel diseases (IBD) and focuses on the role of the desmosomal integrity including the plaqueproteins plakoglobin and plakophillin in the gut. Due to the great clinical relevance of the issue as well as the department structure as department of surgery, the group provides a strong translational component based on the access to a large databank of patient specimens from patients suffering from IBD. The research group develops human and murine 3D and 2D tissue models based on biological scaffolds and primary intestinal organoids combined with cell lines. In addition, in vivo animal studies are performed to further evaluate the findings in a complex microenvironment. As part of a DFG Grant, we recently revealed novel aspects of the role of plakoglobin and plakophillin on barrier function and mucosal healing. Based on those findings, the aim of the PhD project is to develop and characterize the role of the plaqueproteins in vitro with the main focus on organoids as well as in vivo using a mouse model with a selective intestinal knockout of plakoglobin as well as plakophilin. The project will utilize organoid culture as well as advanced molecular and cell biology techniques that are wellembedded at the institute.
Qualification profile:
  • Master or diploma in biology, biomedicine, biochemistry or related fields
  • Strong background/interest in the field of gastro-intestinal and infection biology, immunology, tissue engineering
  • Advanced knowledge in cell culture (optimal: organoid culture), immunohistological staining methods, fluorescence microscopy, molecular biological and biochemical techniques
  • Experience in in vivo work is not necessary but helpful
  • Fluent English, excellent oral and written scientific communication skills
  • High flexibility and the ability to work as part of a team and independently

The position is situated within the DFG Grant SCHL 1962/8-1 "The Role of Plakoglobin on intestinal Wound Healing and Barrier Regulation in Inflammatory Bowel Disease" and offers a 3 years (TV-L) salary. A disabled person will be employed preferably, if qualifications are equal.
